UCI DNA Core provides a comprehensive service of automated DNA sequencing and gene scan analysis for the entire university as well as external customers. The service of automated DNA sequencing currently available is Do-It-Yourself Service. UCI customers should provide the Core a recharge account number upon each order. External customers should have a sales and service account set up in our Core for billing before the service is provided. The DNA sequencing service operated in the UCI DNA Core is on a first-come-first-served basis. However, it is the Cores responsibility to operate the Facility in the most efficient way. Therefore, the operation of service will be prioritized to maximize full plate runs. Do-It-Yourself is coordinated by Core personnel only when it makes sense to do so. Priority of service will also be given to UCI customers.
One pGEM / M-13 Universal primer control will be conducted along with a batch of reaction (CSR) and at least one such control will be run (AGE) along with the corresponding batch of extension products (as well as other GOS extension products) on a gel for FSS and GOS services only. We sometimes use other primers in our primer library for pGEM standard control. This standard control is operated at Core's cost. Any other type of control required by customer, is considered as a service order and will be charged depending on the service type.
Every gel run (AGE) will be checked on its correspoding gel image for quality perposes based on our knowledge and experience of automated DNA sequencing. We will check the gel to see 1) if the gel is running properly, 2) if the gel image is able to be re-analyzed, 3) if the tracking of each lane is correct, 4) if the standard control(s) is good, and 5) if the loading was good and there was no leakage across the wells (377 only). If all the five above-mentioned aspects are positive, such gel run (AGE) as well as the corresponding reaction (CSR) is considered GOOD. If the tracking is incorrect initially but we are able to correct it, it is still considered GOOD. If there is a leakage across the wells but the influence of leakage is irrelevent (at background noise level), it is also considered as GOOD.
